A Parametric Study of Production of Maltotetraose - Producing Amylase in Recombinant Bacillus subtilis LKS88

Abstract
Recombinant Bacillus subtilis LKS88[pASA240] containing the amylase gene from Streptomyces albus KSM-35 was employed for mass production of maltotetraose-producing amylase. The effects of four medium components including phosphate, yeast extract, peptone, and sodium citrate on amylase production were examined in shake flask cultures to determine an optimal medium composition. The optimized medium was verified by experiments in a fermentor and yielded 2.1 and 2.5 times higher cell concentration and amylase activity, respectively, than the standard medium. The optimum pH and temperature for fermentor cultivation were found to be 7.0 and 37¡É. Substitution of glucose with starch as a carbon source resulted in 1.3 times increase in maximum amylase activity, corresponding to 7 times higher amylase production than the standard medium in a shake flask.
